Bone Regeneration Idea | 3D Printed Microfibers Activate the Nerve-Bone Axis

Detailed

Nell-1 promotes osteogenic differentiation and mineralization of hPDLSCs and upregulates CYFIP1, Runx2, and CGRP-related signaling; disrupting CYFIP1 affects the osteogenic effects mediated by Nell-1. The CPC-aMF scaffold has a hierarchically micro-fibrous structure, suitable mechanical properties, and degradation behavior, which can protect the seed cells and achieve dual-phase release of Nell-1. In vivo, scaffolds containing hPDLSCs and Nell-1 promote new bone formation, collagen deposition, and defect repair, while also enhancing angiogenesis and nerve ingrowth, with increased markers like CGRP, β3-tubulin, and CD31. Mechanistically, Nell-1 links nerves and osteogenesis through the CYFIP1-MAPK/ERK-Runx2 axis.
