Despite a lot of efforts, it is still difficult to accurately determine the tomol level of trace biomolecules. Here, Hou Changjun, Luo Yang, Danqun Huo and others from Chongqing University reported a 3D carbon nano-maze (CAM) electrode for ultra-sensitive detection of trace biomolecules, such as nucleic acids, proteins and extracellular vesicles.




Key points of this article:

1) The CAM electrode is composed of a staggered carbon fiber array, and the crossed graphene sheets are vertically bound in situ on it, thereby realizing the local restriction of trace molecules and improving the efficiency of molecular hybridization. In addition, the self-assembled DNA tetrahedral array adopts a rigid spatial conformation to ensure the controllable arrangement of the immobilized biological probes, thereby improving analysis sensitivity and reproducibility.




2) In the proof-of-concept experiment for detecting microRNA-155, linearity from 0.1 aM to 100 nM and sensitivity of 0.023 aM (23 zM) were achieved. Under the optimal parameters, the nanoelectrode has an encouraging consistency with the quantitative real-time polymerase chain reaction in the clinical sample detection process.



In summary, by attaching various interesting biomolecular probes for simple functionalization, the developed CAM platform with ultra-high sensitivity can be used as a general tool in the fields of environment, chemistry, biology and healthcare.
