The clinical management of periodontitis requires addressing both bacterial infection and a bone-deficient microenvironment to optimize tissue regeneration. Electroactive biomaterials have shown potential in bone regeneration, but they are still insufficient to consistently integrate antibacterial properties with optimal immune modulation and tissue remodeling in complex inflammatory microenvironments. This study proposes a highly efficient, dynamic magnetoelectric therapy strategy that combines antibacterial activity with immune regulation to optimize periodontal tissue regeneration.Magnetically sensitive CoFe2O4-BiFeO3 nanoparticles (CFO-BFO NPs) have been developed as a groundbreaking magnetoelectric biomaterial for the treatment of periodontitis. Magnetoelectric CFO-BFO nanoparticles significantly enhance the osteogenic activity of periodontal ligament stem cells (PDLSCs) and exhibit strong antibacterial activity in vitro. Importantly, magnetoelectric coupling stimulation can program macrophages from the M1 to M2 phenotype via the PI3K-Akt signaling pathway, thereby enhancing osteogenesis.In addition, the NPs of magnetoelectric CFO-BFO can alleviate tissue inflammation in a mouse model of periodontitis mediated by Porphyromonas gingivalis (P. gingivalis) and accelerate the regeneration of infected bone. After 4 weeks of treatment, the bone volume fraction (BV/TV) in the magnetoelectric CFO-BFO group approached healthy levels. This study pioneeringly applies magnetoelectric coupling materials to periodontitis treatment and provides a new strategy to enhance the therapeutic effect of electroactive biomaterials on periodontitis.
