Nanozymes have been attracting increasing attention due to their high stability, low cost, and scalable synthesis. However, their clinical potential is often limited by relatively low catalytic efficiency and poor substrate specificity. Recent developments in light-responsive nanozymes offer promising solutions by integrating photocatalytic and photo-assisted mechanisms to enhance activity and spatiotemporal control. These systems improve catalytic performance through light-induced charge separation, generation of reactive oxygen species, and auxiliary photothermal or photodynamic effects.By leveraging these light-responsive mechanisms, various nanozyme systems, including plasmonic metals, metal oxides, semiconductors, and metal-organic frameworks, demonstrate enhanced catalytic functions and outstanding therapeutic effects in cancer treatment, infection control, immune modulation, and tissue repair. This review summarizes the latest advances in the design and application of light-responsive nanozymes and highlights the challenges in clinical translation, positioning them as next-generation strategies for precise biocatalysis and disease intervention.
