Advanced Functional Materials | Biomimetic Hydrogels Prepared from Core-Shell Nanofibers: Accelerating Wound Healing by Modulating Macrophage Polarization

已传文件:photo/1773121782.png Fascia is a hierarchical composite structure within the skin, which can promote rapid repair of damaged tissues and reduce scar formation. However, its significant role in wound healing has been seriously overlooked. Therefore, we developed a wound dressing with a nanofiber network structure, mimicking the skin fascia. Using methylacrylate chondroitin sulfate as the hydrogel substrate, by electro-ionizing loading of bovine serum albumin nanoparticles (BSA @ PFD) onto the dextran shell, a slightly clickable fascia-inspired nanofiber hydrogel (CA-DAF-BSA @ PFD) was constructed, forming a core-shell fiber network. This hydrogel exhibits excellent viscoelasticity, injectability, anti-swelling, antibacterial activity, and biocompatibility. In vitro cell studies demonstrated that the CA-DAF-BSA @ PFD hydrogel can induce macrophages to polarize towards the M2 phenotype and inhibit fibroblast differentiation into myofibroblasts. In vivo, the CA-DAF-BSA @ PFD hydrogel can significantly promote granulation tissue regeneration and effectively reduce scar tissue formation by inhibiting excessive collagen deposition and abnormal fibrosis. This core-shell fiber hydrogel shows great potential as an advanced wound dressing, providing a new therapeutic strategy that can achieve efficient and biocompatible tissue repair.
