After cell lysis, standard procedures can be completed in less than 30 minutes, and the resulting purified DNA is greater than 30 kb. DNA yield varies across species and tissues based on genome size, ploidy, cell number, and age of tissue samples. Typical yields from standard tissue sources (eg, young wheat leaves) are 30 to 32 μg from 100 mg of tissue. The isolated DNA can be used directly for PCR, qPCR, Southern blotting and enzymatic reactions.
