It is a major research challenge to prepare functional components with clear structures through controlled molecular stacking under physiological conditions. Associate Researcher An Hongwei and Researcher Wang Hao of the National Nanoscience Center have modularly designed a type of peptide-anthocyanin conjugate, which can self-assemble into a controllable one-dimensional columnar superstructure in the cell and has unique imaging And thermal performance.







Key points of this article:

(1) The peptide backbone will be cleaved by caspase-3/7 after entering the cell. Subsequently, the self-assembled residues with the bianthocyanidin substitution group (Pr-2Cy) will be arranged to form a P helical column, in which the photothermal conversion efficiency of the H-aggregate of the anthocyanin dye is 3.4 times that of the free dye.






(2) Self-assembled residues (Pr-1Cy) with single anthocyanin substituents will form a loose column structure, in which the fluorescence quantum yield of the anthocyanin dye can reach 9.5% (in H2O, the emission wavelength is 819 nm). In summary, this work provides a new and simple method for functional molecular modification of self-assembling peptides in vivo to achieve their expected biological applications.
